Product Usage Information

This product is the carrier free version of product #57056. All data were generated using the same antibody clone in the standard formulation which contains BSA and glycerol.

This formulation is ideal for use with technologies requiring specialized or custom antibody labeling, including fluorophores, metals, lanthanides, and oligonucleotides. It is not recommended for ChIP, ChIP-seq, CUT&RUN or CUT&Tag assays. If you require a carrier free formulation for chromatin profiling, please contact us. Optimal dilutions/concentrations should be determined by the end user.

Storage

Store at -20°C. This product will freeze at -20°C so it is recommended to aliquot into single-use vials to avoid multiple freeze/thaw cycles. A slight precipitate may be present and can be dissolved by gently vortexing. This will not interfere with antibody performance.

Specificity / Sensitivity

Cathepsin K (E7U5N) Rabbit mAb (BSA and Azide Free) recognizes endogenous levels of total cathepsin K protein. Non-specific focal cytoplasmic staining was observed in mouse testis by immunohistochemistry.

Monoclonal antibody is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Gly230 of mouse cathepsin K protein.

Background

Cathepsin K (CTSK) is a cysteine proteinase belonging to the cathepsin family, which all share a common papain-like structural fold and a conserved active site Cys-Asn-His triad of residues (1-3). Cathepsin K, initially synthesized as an inactive procathepsin K, is further processed into a mature enzyme, whose most studied function is to mediate bone resorption (1-6). Originally discovered in osteoclasts, its expression has also been associated with various cancers, such as prostate, breast, bone, lung, and renal carcinoma (6). Many pharmaceutical companies are working on the development of selective inhibitors for cathepsin K for anti-osteoporosis drugs (5).
