Western blot analysis of extracts from mouse primary bone marrow-derived macrophage (BMDM) cells, untreated (-), or treated (+) with Lipopolysaccharides (LPS) #14011 (50 ng/ml, 4 hr) and Nigericin (sodium salt) (45 min) at the indicated concentrations. Western blot analysis was performed using IL-1β (D3H1Z) Rabbit mAb (Mouse Specific) #12507 (upper), Cleaved-IL-1β (Asp117) Antibody (Mouse Specific) #52718 (middle), or β-Actin (D6A8) Rabbit mAb #8457 (lower).
Confocal immunofluorescent analysis of mouse primary bone marrow-derived macrophages (BMDMs), untreated (left) or treated with Lipopolysaccharides (LPS) #14011 (50 ng/ml, 4 hr) followed by Nigericin (sodium salt) (15 μM, 45 min; right), using ASC/TMS1 (D2W8U) Rabbit mAb (Mouse Specific) #67824 (green). Blue = Hoechst 33342 #4082 (fluorescent DNA dye). Note the translocation of ASC to inflammasomes following stimulation with LPS and Nigericin (sodium salt).

Product Usage Information

Nigericin (sodium salt) is supplied as a lyophilized powder. For a 10 mM stock, reconstitute 5 mg of powder in 669 μl of DMSO. Working concentrations and length of treatment can vary depending on the desired effect.

Storage

Store lyophilized at 4ºC, desiccated. In lyophilized form, the chemical is stable for 24 months. Once in solution, store at -20ºC and use within 2 months to prevent loss of potency. Aliquot to avoid multiple freeze/thaw cycles.

Background

Nigericin (sodium salt) is an electroneutral K+/H+ ionophore derived from S. hygroscopicus. This antibiotic is lipophilic and selectively causes efflux of potassium across mitochondrial membranes (1,2). The ion exchange can lead to disruption of membrane potential, changes in pH, and cell death making the compound useful for studying areas of interests such as apoptosis, mitosis, and the immune response (3).
